Phorbol Ester and Dioctanoylglycerol Stimulate Membrane Association of Protein

چکیده
We used left ventricular myocytes from adult rats to investigate the effect of 4f.-phorbol 12-myristate 13-acetate (PMA) and of sn-1,2-dioctanoylglycerol (DiC-8) on the membrane association of protein kinase C (PKC), cytosolic [Ca2], (Cai) homeostasis, and the contractile properties of single cardiac cells. Because PKC activity is known to be highly Ca2+ sensitive, the K+ concentration of the bathing medium was raised from 5 to 30 mM in some experiments, a perturbation known to depolarize the cell and increase Cai. In cell suspensions both PMA (3 x 10and 3X i0` M) and DiC-8 (10-` and 10-4 M) increased membrane association of PKC. The effect ofPMA (10-` M) on PKC translocation was enhanced in 30 mM KCI compared with 5 mM KCI. During steady field stimulation at 1 Hz in 1 mM bathing [Ca2+], both PMA (10-` M) and DiC-8 (10-` M) decreased twitch amplitude to approximately 60% of control in

Phospholamban and Troponin I Are Substrates for Protein Kinase C In Vitro but Not in Intact Beating Guinea Pig
The incorporation of [32Plinorganic phosphate into membranous, myofibrillar, and cytosolic proteins was studied in Langendorff-perfused guinea pig hearts treated with phorbol 12myristate 13-acetate (PMA) or 1,2-dioctanoylglycerol (D8G), which are potent activators of protein kinase C. Control hearts were perfused with an inactive phorbol ester (4c-phorbol 12,13-didecanoate), which does not caus...
متن کاملInsulin and phorbol ester stimulate conductive Na+ transport through a common pathway.
Insulin stimulates Na+ transport across frog skin, toad urinary bladder, and the distal renal nephron. This stimulation reflects an increase in apical membrane Na+ permeability and a stimulation of the basolateral membrane Na,K-exchange pump. Considerable indirect evidence has suggested that the apical natriferic effect of insulin is mediated by activation of protein kinase C. However, no direc...
متن کاملTumor-promoting phorbol esters inhibit cardiac functions and induce redistribution of protein kinase C in perfused beating rat heart.
Activation of protein kinase C has been implicated in the regulation of a variety of cellular reactions. Although we, and others, have found protein kinase C and its substrate proteins to be present in both membrane and cytosolic fractions in the heart, the physiologic role of this kinase in the regulation of cardiac functions remains unknown. In the present study, we found that in isolated per...
متن کاملPlatelet activation by simultaneous actions of diacylglycerol and unsaturated fatty acids.
Several cis-unsaturated fatty acids such as oleic, linoleic, linolenic, eicosapentaenoic, and docosahexaenoic acids added directly to intact human platelets greatly enhance protein kinase C activation as judged by the phosphorylation of its specific endogenous substrate, a 47-kDa protein. This enhancement absolutely requires the presence of a membrane-permeant diacylglycerol, 1,2-dioctanoylglyc...
